[Abstract] Goal: To study variation pattern and clinic meaning of IFN-y and IL-6 levels produced by the peripheral mononuclear cells (PBMC) in different HBV-DNA load in chronic hepatitis B (CHB) patients before and after Lamivudine treatment. Methods: Blood serum HBV-DNA loads were measured by using fluorescence quantitative polymerase chin reaction (FQ-PCR). IL-6 and IFN-y levels produced by PMBC were measured by using the Double Antibody Sandwich ELISA (DAS-ELISA). Based on the HBV-DNA loads (Copies.ml-1), patients were divided into groups of High-level Viral Loads (>107 Copies.ml-1), Mid-level Viral Loads (105~106 Copies.ml-1) and Low-level Viral Loads (<105 Copies.ml-1). Results: Comparison between CHB patients before treatment: IFN-y was significantly lower (P<0.01), IL-6 was significantly higher (P<0.01). Comparison among High-, Mid-, and Low-level Viral Loads: IFN-y and IL-6 contents were significantly different (P<0.05, P<0.05), with the lowest IFN-y levels in High-level Viral Load group and highest in Low-level Viral Load group. IL-6 levels were lowest in Low-level Viral Load group and highest in High-level Viral Load group. After treatment there was no difference between the control. Conclusion: The difference in HBV-DNA loads in CHB patients after Lamivudine treatment responses to the levels of IFN-y and IL-6. The contents of IFN-y and IL-6 can closed reflect the level of TH1/TH2, and can be used as reference index for CHB treatment.

[Abstract] Objective To investigate the different HBV-DNA load in chronic hepatitis B (CHB) patients treated with lamivudine before and after peripheral blood mononuclear cells (PMBC) secretion of IFN-γ, IL-6 level changes and its clinical significance. Ways of serum HBV-DNA load using fluorescence quantitative polymerase chain reaction (FQ-PCR) determination, PMBC secretion of IL-6, IFN-γ level of the use of double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) determination; by HBVDNA content (Copies.ml-1) to CHB patients were divided into groups of high viral load (> 107 Copies.ml-1), contained in the volume group (106 ~ 105 Copies.ml-1) and low-load group (<105 Copies . ml-1). The results of pre-treatment of patients with CHB compared with normal control group: IFN-γ significantly lower (P <0.01), IL-6 was significantly higher (P <0.01), high, medium and low load comparison between groups: IFN-γ and IL -6 content has significant difference (P <0.05, P <0.05), IFN-γ in the high minimum load group, low-load group of the highest; IL-6 in the low load group the lowest, the highest high-load group. After treatment compared with the control group no difference. Conclusions of different HBV-DNA load in CHB patients treated with lamivudine response and IFN-γ, IL-6 level of the degree, IFN-γ and IL-6 levels to better reflect the level of TH1/TH2 and can be CHB effect as a reference indicator.
